In the absence of any one else, I'l start the ball rolling. Grid completed, apart from the entries for 4 thematic clues, where several words would fit in each case. The work in question doesn't seem to produce a lot of quotations in ODQ, so I have no idea what's happening with the modifications. Hints welcome!

Haven't finished yet, the endgame is eluding me still, but I think I've filled the grid to my satisfaction. Some of the thematic clues are really difficult to untangle. The quotation from the work is pretty famous! So you shouldn't be looking for anything TOO obscure...

It seems to me that in 34Down we could have Cal, Dan, or even Man itself. Not great.

I stumbled across the quote - which is famous, though I had no idea where it came from - by googling the author.

I think I have a full grid though I am struggling to understand 16ac and 24 dn - which I think are probably both thematically modified? Any oblique hints for these would be appreciated.

I've finished all this bar two things: am in the same position as throck for 34d - and I don't see how the final letter of 43ac resolves itself - although in both cases it may depend on solving the original clue which I've failed to do so far

Aspria - I may be wrong but have rest as support and a word for siege equipment inside it for 43 ac. Hope this helps.

24dn is a simple enough clue, be sure to solve it.

16ac was my last one, hardish but if you work out a suitable thematic word for replacement you may find the light dawning as it did for me.

Has anyone sorted out the endgame yet? Still rather baffled there.

Thanks Verlaine, the light has indeed dawned on 16ac. I'm sure I'll get 24d eventually.

Like so many of these rather dreary clues 24d is an anagram.

Having real trouble with the work. Are the initial letters in clue order? (it doesn't say. Can see I've made an error in that regard
