This time I've managed to complete the grid. Thanks to the help on this thread I've located the quote. However... (as ever)...
I'm still stuck on 18 across - where does the whale come in?
I have four cells (in 32a and 14 down) where there are ambiguities. I'm guessing that they should be resolved in some way and resolving the title of the puzzle (an anagram of the title of the work less two letters) should assist.

However I'm stuck, will not enter, am about to watch to football so would welcome any appropriate nudge.

the word missing from the quote is ....

put the 2 letter abbrev for it in front of your jumbles and make real words ....hence no ambiguity on entering

Was going along nicely but, as usual, I’m stuck on two four letter words, namely 28 and 29. Can anyone explain the wordplay please.

28 Singer is the definition. Threaten is surplus. R from racketeers but into an uncomplimentary word for a big city, often used to refer to London.

29 Taught is surplus. A word for "before anything else" losing a traditional way of representing reading, amongst other things, to give a word for handwriting.
