Clue: Quietly stroll around Mile End as an introduction.
I know the answer is PREAMBLE and can see the quietly bit (p) and the stroll (amble), but why is Mile End in the clue?

...and the stroll is RAMBLE rather than AMBLE (though amble would be more accurate!)

The stroll is RAMBLE (not AMBLE), and the idea is that 'Mile End' should indicate the last letter ('End') of the word MILE, ie an E, but why setters think that 'Mile End' = 'the End of Mile' or 'Head Start' = 'the start of head' I have never understood.

...I should have said 'some setters' - many of course would never use 'Mile End' for E.
