DISTRESS - teacher is mistress. Take M (opener) out for D (roman 500) to get DISTRESS (upset).

The clue and your parsing would seem to suggest MISTRESS rather than DISTRESS

Teacher (MISTRESS) become upset (DISTRESS) when opener is out (M) and replaced by 500 (D)?

I've solved 2 in the Wee Stinker and put it aside!
