'The subsiary indication in twelve across clues leads to an extra letter which solvers shouluse to replace a letter in the definition of a down clue ...': i.e., as printed the two down clues that are stumping me have a deliberate misprint in the definition. So the first part of the problem is to identify and definition; the second part is to correct it with any of the following letters which I have identified so far in the across-clues: E, P, F, R, N, W,S, E, T, C, H; but I don't see how that can help much anyway except negatively. Phew.

16D Pashmina
22D Ratter (L out of Rattler and he rots should be he rats)

I've filled the grid but have no idea about replacing certain letters in the grid.

I have the twelve letters 'bumped' from the definitions in the down clues as MLMTPTADOFROM. Which is very little help, as I now have no idea what do with them. Ten of them are supposed to replace symmetrically placed letters in the grid - but where and why? And what to do with the two that are left over?

Dear Cinnabar,
I agree independently with you for the first seven of the letters to be discarded from the down-clues, these being in 2, 3, 5, 7, 8, 9, 10 down respectively: but for the latter five, instead of your O F R O M, I've got D O R M S, finding these letters, in the clues to 20, 22, 25, 28, 29 down respectively. Perhaps worth doing a check of your list? I certainly hesitate to claim I'm right and you;re not; and as for using ten of these to replace letters in ten of the entries symmetrically placed,{u}and generating real words throughout - I'm still at sea...!
All the best

for what its worth I can confirm with cinnabar letters are MLMTPTADOFRM from clues 2,3,5,7,8,9,10,20,22,23,25,28 (cinnabar you seem to have an extra O ) but what now I also have absolutely no idea
