the clue is "Set in" the answer I need is 5 letters long. ?rise if all my other clues are correct.

set in = to begin (among other meanings)
begin = arise. So possibly ARISE

Hi Rambler. If you are looking at the puzzle its easy to see,it just depends on how the blocks are laid out. In this case, the missing letter was at the beginning of the row and the rest filled in from the ones going down.
