It's just that (Job's)Comforter=Job can't be right? Job's Wife is not Job.
"puzzled reflection" could translate to "quizzical thought" ie "mmm, thats odd"

Irritating isn't it. As I understand it the meaning of the phrase "job's comforter" is: someone who while seeming to offer comfort is actually making things worse. They are tactless, malicious or a false friend. So as far as your clue is concerned comforter could well equal job.
What letters do you have that you are sure of?

A new thought. As you pointed out Job isn't the comforter. So, how about NON JOB for the answer. The comforter is definitely not (i.e. non) job. Not much to do is a non job. I suppose strictly speaking it should be hypenated but that doesn't really matter.

I meant to add that puzzled reflection could be explained along the lines of one of those faux french phrases people sometimes use. i.e. moi Job? Non. Surprised (or puzzled) to be asked. And of course,a reflection of the words non job.

has to be odd,job, methinks.
Job (if I remember bible class correctly) rhymes with lobe - as in earlobe. I am drunk now.
ho-hum.

p.s. I never went to bible class.
